Astronauten Dennis Gamble (Mykelti Williamson) en Patrick Ross (Justin Lazard) maken deel uit van de eerste bemande missie naar Mars. Wanneer ze terugkeren op Aarde moeten de overgebleven teamleden uit de eerste film samenwerken met de nu vriendelijke, genetisch gemanipuleerde Sil (Henstridge) om hem te pakken te krijgen.
